1. Summary

In our previous study, we found that many cell lines can form 3D spherical aggregates by using hand casting method, which can mimic the real physiological conditions. Some reports also claims that cell also demonstrates the same behaviour via tuning the adhesive/non-adhesive substrate. In the current study, we prepared the substrate by using cell non-adhesive polymer PEG and cell adhesive protein fibronectin as substrates for the cellular aggregates observation. It concludes that the higher ratio of PEG to fibronectin leads to stronger cell aggregation behaviours.

2. Experimental

Using TIRF and Conforcal laser scanning microscope for the cellular aggregates long-term observation. For each batch of experiment, the different ratio of PEG and fibronectin mixture was used to prepare (70% Fibronectin /30% PEG, 80% Fibronectin /20% PEG, and 100% Fibronectin /0% PEG) as substrates. Then cell aggregates were seeded onto those substrates for 72 hours observations. For each substrate, 10 aggregates were chosen for the observation.

3. Results and Discussion

As a different batches of experiment, we found cell formed different spheroid morphologies in the different PEG/fibronectin ratio prepared substrates. The non-adhesive substrate 70% Fibronectin /30% PEG, cell aggregates maintain its spheroid morphology. However, for the adhesive substrate(100% Fibronectin /0% PEG), the cell aggregates vanished and forms adhesive behaviours.
